BILDIRCINLARDA FARKLI DOZLARDA ORAL UYGULANAN TOLFENAMİK ASİDİN ETKİSİNİN PATOLOJİK OLARAK İNCELENMESİ

Proje Konusu

The aim of this study was to determine the haematological, biochemical and pathological effects of tolfenamic acid (TA) in Japanese quails (Coturnix coturnix japonica) following repeated oral administration of 2 and 8 mg/kg doses. In the study, 42 quails, 60- 70 days old, with an average live weight of 210±15 g, were randomly divided into three groups. The first group (n=6) received saline orally every 12 hours for 7 days. The second (n=18) and third (n=18) groups received 2 and 8 mg/kg TA orally every 12 hours for 7 days at a total of 14 doses, respectively. Haemogram and percentage leukocyte type in whole blood samples and biochemistry parameters in serum samples were determined. Liver, kidney and spleen tissues were taken for histopathological examination. When the study groups were compared, other haematological parameters except MCHC were similar between the groups. MCHC value was lower in the 8 mg/kg dose group compared to the control group. In the 8 mg/kg dose group, basophil and monocyte percentages were higher and lower, respectively, than in the 2 mg/kg dose group. Administration of TA at different doses did not change the biochemical parameters. There was no statistically significant difference between the groups in terms of relative liver, kidney and spleen weights (P>0.05). It was observed that the livers were paler in 2 mg/kg and 8 mg/kg groups (moderate and severe) compared to the control group and a statistically significant difference was observed between the control group and the experimental groups (P<0.05). A statistically significant difference was observed between the control group and 2 mg/kg and 8 mg/kg groups in terms of hydropic degeneration/ lipidosis in the liver, degeneration in the tubule epithelium in the kidney and decrease in lymphoid tissue in the spleen (P<0.05). The results of the study showed that TA 2 and 8 mg/kg doses administered orally every 12 hours for 7 days may cause hepatotoxic and nephrotoxic effects without causing changes in haemogram and biochemical clinical parameters. Therefore, molecular studies are needed to determine the mechanisms of hepatotoxic and nephrotoxic effects of TA in quail.
